He lives with his older sister, Tae Shimura, at their family's sword dojo, the Tendo Mushin-ryu. First person: I (polite). He usually speaks formally, but occasionally uses "I" (casual/masculine). In a series full of intense eccentricities, he is one of the few sane straight-men. He serves as the narrator and the story's observer, effectively the second protagonist with the most appearances after Gintoki... yet he remains a plain boy with a weak presence. According to the author, Shinpachi's design is the base for all other characters. He is famous for his "OI!!!" style shouting retorts. He is teased so much for being a plain guy with glasses that people say the glasses are his actual body.

She usually speaks in broken Japanese, ending sentences with "-aru," "-ne," "-yo," or "-yoroshi" (stereotypical Chinese-accented Japanese), though she occasionally uses standard Japanese or Kansai dialect for jokes. Due to the nature of the gag manga, she often ends up in gross situations; despite being the heroine, she has vomited on screen, earning her the nickname "Geroine" (Vomit-heroine). She wears a cheongsam. As a member of the Yato tribe, she has translucent white skin, light blue or purple eyes, and salmon-pink hair tied into two buns with ornaments. He is a heavy smoker and a severe mayonnaise addict. He was once possessed by the cursed sword "Muramasha," giving birth to a second personality named "Tosshi." As the Shinsengumi's number two, he is known as the "Brains of the Shinsengumi." He is the most popular man in the force and is handsome, though his pupils are always dilated. Unlike Gintoki, he is often drawn without highlights in his eyes. He is extremely proud, serious, and cool. He is a relatively sane character who handles the straight-man role. While he has a blunt and stubborn attitude, he is actually caring and shows a clumsy kind of kindness. Because he deals with problem children both above and below him, he is great at following up, earning the nickname "Follow-kata Toshi-follow." Despite his serious nature, he rarely uses honorifics with superiors, though he uses polite speech with Tokugawa Shigeshige or students on field trips. He is a die-hard Nintendo fan and a Mario lover. Also, despite being in Jump, he is a devoted reader of Weekly Shonen Magazine and loves old-school delinquent manga, even making a rule that no manga other than Magazine is allowed in the station. He is one of the few in the Shinsengumi who is actually hygienic, such as washing his hands after using the toilet.

He has a cute baby face, but in short, he is "black-hearted, sharp-tongued, and a total sadist." He is constantly aiming for the Vice-Commander position and Hijikata's life. Because of his sadism, Hijikata calls him the "Prince from the Planet Sadistic," and he has been labeled the "Sado-Prince" in newspapers. He speaks in a low-energy, detached tone. He is quick-witted and has even tried to trick the readers; he has an eccentric personality that goes beyond just being a sadist. Despite being a minor, he is depicted as enjoying alcohol. He is a chronic slacker with a lazy work ethic, which earns him the resentment of the serious Hijikata. He speaks with a constant, weary Edo dialect. He loves listening to Rakugo programs. When napping, he wears a red eye mask with eyes drawn on it. He is skilled at most things and supposedly holds a "Space Toxic and Hazardous Substances Handling License" in addition to a driver's license. Like Katsura, he can sleep with his eyes open and even has the talent to sleep while standing.
