Casey Jones is corrections officer turned hockey-masked vigilante. Casey Jones starts the movie as a stubborn corrections officer transporting the Shredder. When the Foot Clan hijacks the convoy, he is fired and decides to take justice into his own hands using sports gear. He met April O'Neil when he saved her from attackers

April O'Neil serves as an investigative ally to the Turtles, tipping them off to Shredder's ideas and hiding spots. She is an ally to the Turtles. April acts as the Turtles' eyes and ears on the surface, feeding them information and stepping in to divert attention when they are cornered.

Shredder is the supreme leader of the Foot Clan and he escapes police custody with the help of scientist Baxter Stockman to unleash the alien warlord Krang. Shredder returns to exact his revenge. He orchestrates a prison break for revenge. Unlike the large, mechanized metallic suit from the first film, this version of Shredder sports a darker, more traditional samurai-inspired armor.

Baxter Stockman is a brilliant, sociopathic mad scientist working for TCRI who initially aids the Foot Clan in an attempt to break Shredder out of prison. Baxter engineers a teleportation device to free Shredder during a prison transport, though the turtles ultimately interfere. He uses a purple ooze also known at mutagen and makes more of it for shredder
