Lupin III steals the Philosopher's Stone, rumored to grant "eternal life," from a pyramid in Egypt. However, immediately afterward, Fujiko Mine snatches the stone and flees to her mysterious client, Mamo.
Around that time, news that Lupin has been executed was shocking the world. However, the executed individual looked exactly like Lupin himself, leading Lupin to question his own existence. After reuniting with Daisuke Jigen and Goemon Ishikawa, Lupin learns that Mamo is the one behind the incident.
Mamo, while posing as the world's wealthiest man, Howard Lockwood, was actually a scientist who had lived for ten thousand years through cloning technology. He called himself "God" and a "Prophet," seeking to achieve perfect immortality using the Philosopher's Stone. Furthermore, he took a liking to Fujiko and intended to keep her by his side.
Mamo used Fujiko to make Lupin steal not only the Philosopher's Stone but also various items related to immortality. However, Lupin realized they were being used and handed over a fake stone. Enraged, Mamo used Flinch and his other subordinates to pursue Lupin's gang, destroying their hideout and capturing Lupin.
Meanwhile, the U.S. government, under threat from Mamo, ordered Special Assistant Stuckey and his subordinate Gordon to handle the situation. Stuckey and his men detained Jigen and Goemon to extract information about Mamo, but eventually began an attack on Mamo's stronghold. Inspector Zenigata was also ordered by his organization to cease his investigation, but he prepared to resign and continued to pursue Lupin alone.
Lupin infiltrated Mamo's base and thwarted a global attack via nuclear missiles. In a final move, Mamo placed his true self—a giant brain—into a capsule and attempted to escape into space via rocket. However, the rocket was destroyed by a bomb Lupin had planted in advance, ending Mamo's ten-thousand-year life. After the incident, Lupin and Zenigata resumed their endless cycle of pursuit and escape.
She betrayed the rest of the Lupin gang, including Guest, to team up with Mamo.
